Abstract
This paper describes the development of a unique fluorescence based DNA diagnostic assay that does not require pre-labeling of the target oligomer prior to analysis. The assay uses polyacrylamide hydrogel plugs immobilized in microfluidic channels, and is based on the displacement of a short sacrificial fluorescent-tagged indicator oligomer by a longer untagged target sequence of interest as it is electrophoresed through the hydrogel plug sensing platform. The distinct advantages of this assay are the short sensing times as a result of directed electrophoretic transport of target DNA to the sensing element and the ability to detect non-labeled target DNA.
