Precise Characterization of Liposome Populations Formed in a 5-Channel Flow-Focused System

Abstract

Previously, we demonstrated liposome formation using hydrodynamic focusing in a microfluidic network which enables us to produce liposomes of a controlled size and size distribution in a reproducible fashion [1]. Former measurements on liposomes were conducted with batch dynamic light scattering (DLS) and multi angle laser light scattering (MALLS) of unfractionated samples. However, this measurement only provides ensemble averages. In this paper, a more detailed and accurate measurement technique that allows for precise characterization of liposome populations using asymmetric flow field flow fractionation (AFFFF) before MALLS and DLS is shown. Details about liposome size and size distribution allow further elucidation about the liposome self-assembly process.
