Focal Adhesion of Osteoblasts-Like Cells on Films of Poly(d,l-lactide)/Poly(vinyl alcohol) Blends

Abstract

Films of poly(d,l-lactic acid) [PLA] and poly(vinyl alcohol) [PVOH] blends were prepared by a solvent casting method with the use of hexafluoroisopropanol (HFIP) as the solvent. The miscibility and the biocompatibility of the blend films over a wide range of composition were studied. Infrared spectra of the blend films that were quenched from 200 oC showed a shift in the carbonyl peak of PLA from 1755 cm-1 to 1731 cm-1 when the compositions of the blend films were not more than 30 % in mass fraction of PLA. Immunofluorescence staining of MC3T3-E1 osteoblast-like cells on blend films was used to evaluate focal adhesions and cell-spread area as markers for biocompatibility.The results showed that the cells seeded on the blend films of intermediate compositions [(55, 70, and 80) % in mass fraction of PLA] exhibited diminished cell-spread areas compared with the cells seeded on the blend films of other composition. Similarly, the focal adhesions of the cells that were seeded on the blend films of intermediate compositions [(70 and 80) % in mass fraction of PLA] were less than the focal adhesions of the cells seeded on the blend films of other compositions, except the blend films that were 55 % in mass fraction of PLA. Thus, focal adhesions and cell-spread areas can be used as indicators of the biocompatibility of PLA/PVOH blends.
