Photopatterned Polyelectrolyte Multilayers for Cell Culture

Abstract
Patterned two-dimensional cell cultures are increasingly important for constructing bioanalytical tools for cell and molecular biology to address questions about interactions, signaling, migration, and differentiation, among others. This contribution presents a unique method for patterning layer-by- layer polymer with dissimilar cell attachment characteristics. Polyelectrolyte multilayers (PEMs) were deposited and exposed with UV light through a binary photomask to form a patterned upper crosslinked stratum where cells are not viable and a lower cytophilic stratum. Specifically, bilayers of polyacrylic acid and polyacrylamide (PAA/PAAm) were patterned (i.e., crosslinked) on top of electrostatically bound layers of biocompatible poly(allylamine hydrochloride)/poly(sodium 4-styrenesulfonate (PAH/PSS) bilayers. This work demonstrates the facile patterning of PEMs for two-dimensional cell cultures that exhibit natural phenotypic expression and long-term viability, which are significant advantages over many other cell patterning strategies.
