Abstract
During the study of Y haplotype distribution of the Short Tandem Repeat (STR) loci included in the commercial kits PowerPlex Y System (Promega Corporation) and AmpFlSTR Yfiler PCR Amplification Kit (Applied Biosystems) we sampled 110 individuals across Central-Southern Italy. We observed off-ladder alleles at two STRs loci (DYS19 and DYS439). We also observed duplications at four loci: DYS439, DYS389II, DYS393 and DYS385. The off-ladder alleles were isolated, and independently sequenced to confirm their designations as: allele 9 from locus DYS19 (two different samples) and allele 7 from locus DYS439.
