Abstract

The use of in vivo transplantation assays under defined experimental conditions has become a valuable standard for evaluating osteogenic potential of different stem cell populations and is the subject of ongoing pre-clinical reconstructive therapy research. Therefore, development of a cost effective in vivo transplantation assay technique offering low volume sampling and rapid analysis in a high-throughput manner is of great interest due to the escalating health care costs. Herein, we have successfully validated an in vivo combinatorial transplant model by developing a miniature cassette transplant with multiple slots for screening osteogenesis of various human bone marrow stromal cells (hBMSCs) and bone grafting formulations (e.g., HA/TCP powder, Gelfoam) in subcutaneous space of immunodeficient mice. Our results confirmed that microwell-type combinatorial cassette transplant model can be used in in vivo cell based transplantation assay applications to efficiently, systematically and rapidly test as many as hBMSCs/vehicles combinations for maximizing osteogenesis.
