The thermodynamic principles that pertain to both chemical and overall biochemical reactions are discussed with particular emphasis on enzyme-catalyzed reactions. Chemical reactions involve specific species (which may also be biochemical importance) and overall biochemical reactions involve substances where one or more of the reactants is a mixture of species. The general principles involved in both equilibrium and calorimetric measurements are discussed with particular emphasis on isothermal microcalorimetry where two solutions are mixed in a reaction vessel. The status of the scientific literature on the thermodynamics of enzyme-catalyzed reactions is summarized along with a discussion of the importance of thermodynamic network calculations on biochemical substances and reactions. The combined use of equilibrium and calorimetric measurements serves as an important tool for elucidating the thermodynamics of the most complex biochemical reactions.
