Fabrication of Polymeric Nanoparticles by Dispersion Polymerization: Review and Data on Stealth PLA-based Nanoparticles

Abstract

Factors influencing size and other characteristics of nanoparticles prepared by free radical dispersion polymerization are discussed. Further, the results of optimization studies designed to minimize the size of crosslinked biodegradable stealth polymeric nanoparticles fabricated in our laboratory are presented. Nanoparticles were fabricated using different amounts of macromonomer, initiators, crosslinking agent and stabilizer in a dioxane/DMSO/water solvent system. Confirmation of nanoparticle formation was achieved by scanning electron microscopy (SEM). Particle size was determined by dynamic light scattering (DLS). D-optimal mixture statistical experimental design was used for model generation (Scheffe polynomial) and optimization. Model verification was done by comparing particle size data of some suggested solutions to the predicted particle sizes. Results show that average particle sizes follow the same trend as those predicted by the model. Negative terms in the model corresponding to the crosslinking agent and stabilizer are important for minimizing particle size.
