Preparing Spectral Libraries of Chloroplast Components for a Raman Imaging and Microspectroscopy Study

Abstract

Raman imaging is a non-invasive vibrational spectroscopic technique that utilizes both Raman spectroscopy and optical microscopy. This technique allows one to simultaneously identify components of biological systems both spectrally and spatially. The unique spectral characteristics of various components will be utilized to identify the ultra-structural regions of plant chloroplasts and their responses to excessive ozone in the presence and absence of an anti-ozonant. A spatial resolution on the order of 1mm and a spectral resolution on the order of 7-8 wavenumbers (cm-1) make this an appropriate method for studying these systems. Among the issues addressed by agricultural biotechnology, the need for sustainable agriculture and improved nutritional quality dictates that we understand and develop methods that effectively and expediently assess the effects of challenging environments on plant tissues. A systematic approach to developing spectral libraries for this Raman imaging study will be presented and used to ascertain whether Raman imaging is a viable measurement tool that provides an alternative to the typical bioassay that describes the effects of environmental stress on plant tissues.
