I specialize in analytical characterization of biomolecules, with an emphasis on separations science and mass spectrometry.  My doctoral work focused on developing peptide-based kinase activity assays in individual human cells using capillary electrophoresis.  In my post-doctoral work, I develop capillary electrophoresis and mass spectrometry assays for structure-function determination of biomolecules.  My work has been instrumental in development and release of NIST’s first monoclonal antibody reference material (NISTmAb RM 8671).  I am currently engaged in designing and implementing a bioassay program to support NIST’s monoclonal antibody reference material program.  Over the course of my career I have become familiar with a variety of analytical techniques including capillary electrophoresis, HPLC, ESI, MALDI, Orbitrap and TOF mass spectrometry, fluorescence, UV/Vis, microscopy, and in-vitro bioassays.

Dr. Mark Lowenthal joined NIST in 2007 following a post-doctoral position at the University of Colorado Health Sciences Center and a research fellowship at the National Cancer Institute (NCI/NIH). Mark received a Ph.D. from the University of Maryland in analytical/physical chemistry and a B.S. from James Madison University.

His research activities at NIST focus on the measurement of proteins, metabolites, amino acids, and sugars in all types of biomaterials.  Current aims include the development of a glycan-based Standard Reference Material, measurement assays for hemoglobin A1c, and creating tools for predicting protein glycosylation using evolutionary sequence conservation. 

Mark has developed new methods for quantification of troponin I in serum - an important protein marker of cardiac muscle injury - using immunoaffinity techniques and mass spectrometry.  Quantification of clinically-relevant biomolecules has led to the production and certification of several NIST Standard Reference Materials.  Other research applications include mass spectrometry-related measurements of proteins associated with neurodegenerative disorders, characterization of antibody function on nanoparticles, and detection of microbial virulence.  

Mark has co-authored 33 peer-reviewed publications and two book chapters.  He is a member of the IFCC CSF working group, GBSC, and the NIH GlycoSIG working group.

David's research at NIST has focused on improving quantitative measurement of proteins, employing both chromatography and mass spectrometry. For the past 15 years, his efforts have been directed at standardizing clinical diagnostic measurement of proteins through the development of certified reference materials and reference measurement procedures. He has collaborated on several standardization projects with the American Association for Clinical Chemistry and the International Federation of Clinical Chemistry and served on committees and working groups for the Clinical and Laboratory Standards Institute. His recent standardization projects include the development of reference materials and measurement procedures for the clinical measurements of cardiac troponin I, C-reactive protein, urine albumin, and prostate specific antigen. Additionally, he has worked in the past three years with the National Cancer Institute to improve measurement quality in clinical proteomic investigations, supporting the Clinical Proteomic Technology Assessment for Cancer (CPTAC) interlaboratory programs.
