Abstract

Localizing a fluorescent particle by scanning a focused laser in its vicinity and analyzing the detected photon stream provides real-time information for a modern class of feedback control systems for particle tracking and trapping. We present a simple mathematical test to determine whether a particular laser scan path encodes maximal information about a fluorescent particle's position in the detected photon stream. We give the optimal path for a Gaussian laser scanned in two or three dimensions, and present the Cram\'{e}r-Rao bound on the two and three-dimensional localization accuracy that can be achieved by analyzing the detected photon stream.
