Synthesis and Characterization of PEG and PEG Urethane Dimethacrylate Hydrogels

Abstract

A key area in the repair and regeneration of tissues is optimizing the polymeric scaffold-tissue response. This study is designated to better understand the relationships between polymer matrix structure and properties to cell response. The current study includes the preparation/characterization of a series of polyethylen glycol (PEG) dimethacrylate, their conversions in aqueous solution to photopolymerization, and a preliminary assessment of the correlation of mechanical and cell response to hydrogel structural variations. PEGs with various molecular masses (Mn=1000 to 8000 e/mol) were reached with methacrylic anhydride or with 2-isocyanatoethyl methacrylate of obtain PEG-dimethacrylate (PEGDM) and PEG-urethane-dimethacylate (PEGUDM), respectively.MALDI-TOF MS confirms the formation of PEGDM oligomers of high purity and narrow distribution (PD<1.02). Aqueous solutions of these dimethacrylates (10% by mass fraction in water) were photopolymerized to hydrogels. Bovine chrondrocytes, seeded in hydrogels, were used to asses the cell responses tp the hydrogels. Preliminary studies showed varied mechanical response but that cells wre completely viable in both types of hydrogels after two weeks. Hydrogels prepared from these dimethacrylates can provide a basis for the understanding how their material properties influence the cell response.
